I am comt ++ and mthfr++, I cannot tolerate even a sliver of Sam'e or many other supplements that help with methylation. I would like to support my methylation cycle. When I take anything with Sam'e I get a headache and feel almost drunk for a few days.

Wait a minute on the Deplin! Use great caution. Deplin is 15mg of methyltetrahydroxyfolate. That's wayyyy too much folate when suggested starting dose is in the mcg's. Psychiatrists are prescribing this indiscriminately due to the on-the-bandwagon approach to epigenetic medicine when they have absolutely no idea what it's all about except what big pharma tells them. Maybe it works for some but it made me absolutely wiggy.

mthfr can be treated with a food prescription called deplin (generic is l-methylfolate). mthfr prevents folate from converting properly to the bio-available form of folate that can cross the blood-brain barrier. L-methylfolate is the bio-available form. A doctor can prescribe it.

It means your system is storing and not utilizing therefore there is other work that needs to be done with your health, maybe detox or maybe supplement deficiencies.
Have the spectacle test done ignored to determine your deficiencies.

It may be that there is another component that is in opposition of that pathway. Maybe starting very slow on methylfolate or methylcobalamin, would be helpful?
